Obinutuzumab (GA101)
GA101 is a type II, glycoengineered, humanized anti-CD20 monoclonal antibody designed to bind with high affinity and selectivity to the extracellular domain of CD20 on the surface of B cells. Binding of GA101 leads to higher cell death induction in malignancies with CD20 positive B-cells.1-3 This agent is being investigated in collaboration with Biogen Idec.
By virtue of its type II antibody characteristics, GA101 causes higher direct cell death induction of cancer cells and lower complement recruitment compared with type I anti-CD20 antibodies.2 Modification of sugars to the Fc region of GA101 (GlycoMAb™ technology) has resulted in stronger binding to immune-effector cells, which translates to enhanced ADCC.2,3
Preclinical studies demonstrate that GA101 induces significant B-cell depletion in peripheral blood as well as in lymphoid tissue. It also mediates high antitumor activity in preclinical NHL studies, both as a single agent and in conjunction with chemotherapy.1
- GA101 is the first type II glycoengineered anti-CD20 monoclonal antibody.1,2

- Preclinical studies have shown that because of its type II mode of binding to the extracellular domain of CD20, GA101 substantially increases direct cell death induction.2

- Glycoengineering of the GA101 molecule enables it to bind with higher affinity to immune effector cells, such as natural killer cells, which bind and target the tumor cells for destruction.3
